Analysis of Coliform Bacteria in PAMSIMAS Drinking Water Samples in Kuningan Regency

Abstract
The increasing activity of the people of Kuningan Regency without paying attention to the environment causes pollution, one of which is pollution of drinking water sources, until reducing its quality. Coliform bacteria are microorganisms that are commonly used as an indicator of water contaminated by pathogens. This study aims to determine the feasibility and the number of Coliform bacteria in PAMSIMAS drinking water samples from seven springs in Kuningan Regency. The analytical method used is based on the Most Probable Number (MPN) method involving two test stages, namely the presumptive test on Single Strength and Triple Strength Lactose Broth (LB) media and Escherichia coli Broth (ECB) media. Then, the confirmation test was continued on Brilliant Green Lactose Broth (BGLB) and Eosin Methylene Blue (EMB) media. The results showed that in the presumptive test all samples in LB media were positive for Coliform bacteria, while in ECB media there were three positive samples containing Escherichia coli bacteria. The confirmation test stage on BGLB media showed that six samples contained different amounts of Coliform bacteria. While the confirmation test on EMB media only one sample which showed a positive result containing E. coli bacteria. Based on the PERMENKES number 492 of 2010 that the PAMSIMAS drinking water samples from seven springs in Kuningan regencyt didn’t meet the quality standards and were not suitable for consumption.
